Sailboat tours. We went on a sunset wine tasting (they gave us a shit ton to taste) and a kayak/snorkel/lunch one. I'll try to find the info for the company we used, they were #1 on trip advisor when we did it. Not cheap, but we'll worth it (especially since my parents paid).
We did the sunset cruise too. The Western Union one I think. They had a live singer on board as entertainment and it was so cheesy but DH bought his CD because it was an awesome cheesy.

Go parasailing, I did with Sebago had an amazing time! Go bar hopping and shopping on Duvall St! There is also a swim with dolphins, dolphins plus i believe was in key largo most definitely worth the trip!!
I took my mom there in October for a kid-free week and it was glorious! I hadn't been there since I was a kid. I definitely second the sailboat tours and Mallory Square is always a blast. We went to the Key West Historical Museum (by Hemmingway's house) and it was really interesting to learn about the history of the island. I'm jealous -- have so much fun!

Also, everything @pobrecita said. We had a "rehearsal lunch" the day before our wedding at Louie's Backyard. Also, it's a little touristy but there is a Cuban place I love near Mallory Square. El Maison de Pepe. There is a place with better Cuban food (according to reviews) but it can't match the location and atmosphere.
Renting bicycles was fun, too. That was our mode of transportation when we rented a house for the week.
The keys are my second home. Dd has been there 15ish times in her life of 3 years. If you plan on having a car and going to other islands I can help with that. I need to get back there in the next couple months.

YES!! Danger Cruises sunset wine tasting is so much fun.
We did a dolphin cruise/snorkel trip. It was so cool; we had a few dolphins swim up along with our boat, including a mother and her nursing calf. We also saw one flinging fish into the air before s/he ate them. It was neat to see them doing their thing in "the wild".
